Abstract

Aging is increasingly viewed as an organism-wide process marked by systemic decline and multimorbidity. This Review frames immunosenescence as a context-dependent mediator, amplifier, or consequence of multi-organ dysfunction, integrating niche-centered mechanisms, bidirectional immune-organ interactions, innate-adaptive remodeling, immune-tissue axes, and multidimensional biomarkers. We also highlight current limitations, including context heterogeneity and unresolved causality, and discuss senolytic, senomorphic, and other therapeutic strategies with potential relevance and limitations for healthspan extension.

The following analysis is generated by AI and may contain errors. It is separate from the authors’ abstract and is not peer review.

Research summary

This review synthesizes current understanding of how immunosenescence drives multi-organ dysfunction and evaluates emerging therapeutic strategies like senolytics, providing a consolidated framework for the field without presenting new primary experimental data or a transformative mechanistic breakthrough. The paper is relevant because it addresses the root causes of aging by focusing on systemic immune decline and its role in multimorbidity, rather than treating isolated symptoms, although its impact is limited by its nature as a narrative review.
